Electrophysiology

Electrophysiology provides a different approach to monitor neuron-to-neuron communication. Neuronal activity is examined by means of electrical recording of action potentials. Ionchannel opening and closing affect the membrane potential (-30 – -90 mV), and are involved in the generation and propagation of action potentials in neurons.

At Brains On-Line we carry out IN VIVO single-cell recordings to evaluate the effects of different pharmaceuticals on:

  • Neuronal firing rate
  • Number of neurons per trackt
  • Long Term Potentiation

Neurons are identified by

  • Their location
  • Waveform
  • Firing rate
  • Response to known (ant)agonists

On this basis we routinely study

  • Serotonergic neurons (DRN/MRN)
  • Adrenergic neurons (LC)
  • Dopaminergic neurons (VTA/SN)
  • Histaminergic neurons (TMN)
  • Glutamatergic neurons

Animals are fixed in position and kept under propofol anesthesia and at constant body temperature; drugs are systemically administered through a lateral tail vein, signal is recorded and analysed with the SPIKE2 software package.
